Ribosomal Crystallography and Heteropolytungstates

چکیده
Heteropolytungstates play a dual role in ribosomal crystallography. Beside generating phases, one of them, was found to be extremely useful in inducing post crystallization rearrangements. These led to a significant increase in the internal order of crystals of the small ribosomal subunits from Thermus thermophilus, manifested in a dramatic extension of the resolution of their diffraction patterns, from the initial 7-9 Å to 3 Å. The current 3.3 Å electron density map of this particle, constructed using phases obtained from this W cluster together with other metal compounds, shows the recognizable overall morphology of the small ribosomal subunit. Over 96% of the nucleotides were traced and the fold of all proteins was determined fully or partially. Specific sites were determined independently by covalently bound heavy atom clusters, among them the surface of two proteins and a functional center, the gate for mRNA binding. All tungsten-cluster sites detected in this map are located in close proximity to the proteins of the particle, in positions that may have an influence on the stability and the rigidity of this rather flexible ribosomal subunit.

The linkage between ribosomal crystallography, metal ions, heteropolytungstates and functional flexibility.
Crystallography of ribosomes, the universal cell nucleoprotein assemblies facilitating the translation of the genetic-code into proteins, met with severe problems owing to their large size, complex structure, inherent flexibility and high conformational variability. For the case of the small ribosomal subunit, which caused extreme difficulties, post crystallization treatment by minute amounts o...

The reaction between [XW(9)O(34)](9-) (X = As, P) and [RuC(6)H(6)Cl(2)](2) in aqueous buffer solution (pH 6.0) leads to the isolation of two pseudo-sandwich-type heteropolytungstates KNa(6)[(RuC(6)H(6))AsW(9)O(34)] x 17 H(2)O (As-1) and Na(7)[(RuC(6)H(6))PW(9)O(34)] x 14 H(2)O (P-2). Single-crystal X-ray diffraction analyses reveal that the two compounds have the following features: (1) in two ...
متن کامل182tungsten Mössbauer spectroscopy of heteropolytungstates.
The tungsten-182 Mössbauer spectra of a series of Keggin structure heteropolytungstates, [EW12O40]n- are reported. There is a very considerable variation in quadrupole coupling at the tungsten nucleus indicating considerable asymmetry in the electron distribution for the more electronegative elements E. The quadrupole coupling correlates well with the structural data, in particular with the dis...
متن کاملReproducible growth of well diffracting ribosomal crystals.
The crystallization of ribosomal particles is associated with extraordinary challenging demands. This originates mainly from the ribosome's natural tendency to deteriorate and from its multi-conformational heterogeneity, both of which stem from its functional flexibility. To increase the level of homogeneity of ribosomal preparations, systematic searches for conditions yielding populations of f...
متن کاملRibosomal Crystallography: Dynamics, Flexibility and Peptide Bond Formation
High-resolution crystal structures of functionally active ribosomal particles provide unique tools for understanding key questions concerning ribosomal function, mobility, dynamics, and involvement in cellular regulation. Structure analysis of complexes of ribosomal particles with substrate analogs and universal drugs indicated that ribosomes provide the structural frame for precise positioning...
